Cellular uptake machinery is a broad term encompassing the various biological pathways and protein complexes that facilitate the internalization of extracellular substances into a cell (Doherty & McMahon, 2009, Annual Review of Biochemistry). These mechanisms include clathrin-mediated endocytosis, caveolae-mediated endocytosis, macropinocytosis, and phagocytosis, as well as specific transmembrane transporters. This machinery is essential for maintaining cellular homeostasis by regulating nutrient uptake, signal transduction, and the recycling of cell surface receptors (Sorkin & von Zastrow, 2009, Nature Reviews Molecular Cell Biology). In clinical contexts, these pathways are often hijacked by pathogens, such as viruses and bacteria, to gain entry into host cells (Mercer et al., 2010, Annual Review of Biochemistry). Additionally, cancer cells frequently exploit these mechanisms to increase the uptake of growth factors and nutrients required for rapid proliferation. While specific components like dynamin or clathrin are studied as potential targets, the ubiquity and essential nature of cellular uptake processes present significant challenges for drug development, primarily due to the high risk of systemic toxicity and off-target effects (Dutta & Donaldson, 2012, Cell Logistics).
