Overview

Cementoblast differentiation is a complex biological process central to the development of the tooth root and the regeneration of the periodontium. It involves the transition of undifferentiated mesenchymal progenitor cells, primarily from the dental follicle or periodontal ligament (PDL), into specialized cementoblasts. These cells are responsible for secreting and mineralizing the cementum, a critical tissue that anchors periodontal ligament fibers to the tooth root surface. The process is tightly regulated by specific molecules such as Cementum Protein 1 (CEMP1) and Cementum Attachment Protein (CAP), as well as transcription factors like Runx2 and Osterix. Signaling pathways including BMP, Wnt, and Hedgehog also play significant roles in modulating this cellular event. Therapeutic strategies for periodontal disease and root resorption frequently aim to stimulate this process using growth factors such as BMP-2 and BMP-7 or enamel matrix derivatives to achieve functional tissue restoration.
