Overview

Adenosine receptors in the central nervous system (CNS) are a family of G protein-coupled receptors (GPCRs) that mediate the physiological and pharmacological effects of adenosine, a purine nucleoside involved in neuromodulation. These receptors play crucial roles in regulating neurotransmitter release, neuronal excitability, synaptic plasticity, and neuroprotection. There are four known subtypes: A1, A2A, A2B, and A3. The most prominent subtypes in the CNS are A1 and A2A. Selective ligands targeting these receptors are under clinical investigation or development for neurological diseases due to their broad regulatory roles within neural circuits.
