Centriolin (CNTRL) is a centrosomal protein essential for centrosome function as a microtubule organizing center in human cells. It plays key roles in mitotic cell cycle progression, particularly during cytokinesis, where it anchors exocyst and SNARE complexes at the midbody to facilitate secretory vesicle-mediated abscission, the final stage of cell division. Centriolin localizes to the centrioles and is involved in centrosome maturation, spindle pole organization, and proper asymmetric division in oocytes, but unlike in mitotic cells, it is not required for cytokinesis in meiotic oocytes. Genomic alterations and misregulation of centriolin have been associated with several diseases, including myeloproliferative disorders and certain cancers due to chromosomal rearrangements involving the CNTRL gene. There are currently no known drugs directly targeting centriolin, and it is not recognized as a standard therapeutic target.
