Overview

Centromere protein B (CENP-B) is an **80 kDa DNA-binding protein** localized to the central region of the centromere in human cells[1][8]. It is highly conserved among mammals and specifically recognizes a 17 base pair DNA motif (the **CENP-B box**) within alpha-satellite repeats of centromeric DNA[1][2][5]. CENP-B contains a **helix-turn-helix DNA-binding domain** in its N-terminal region, which allows it to bind specifically to DNA; it also forms dimers or oligomers through its internal and C-terminal self-association domains, promoting chromatin organization and heterochromatin condensation[3][5][7]. CENP-B contributes to **centromere structure formation**, kinetochore assembly, and centromere stability, working in concert with other centromeric proteins such as CENP-A[9]. It is not essential for basic centromere function, but is required for de novo centromere formation and for stabilizing centromeric architecture[9]. Clinically, CENP-B is recognized as a **major centromere autoantigen**, frequently targeted by autoantibodies in systemic autoimmune diseases and investigated as a **potential cancer biomarker**[1]. No drugs targeting CENP-B are currently known, and it is not considered a direct therapeutic target at this time.
