Centromere protein H (CENP-H) is a centromere and kinetochore-associated protein encoded by the CENPH gene in humans and other mammals[6][8]. It is a key component of the CENPA-NAC (nucleosome-associated) complex, which is essential for the assembly of kinetochore proteins and the integrity of chromosome segregation during mitosis[4][8]. CENP-H forms multiprotein complexes at the inner kinetochore plate, colocalizing with other core centromere proteins such as CENP-A and CENP-C[6][8]. It is constitutively present at kinetochores throughout the cell cycle and is required for proper localization of CENP-C and for chromosome congression and segregation[2][6][8]. The CENP-H protein contains a coiled-coil domain and a nuclear localization signal, suggesting its role in centromere targeting and stabilization[2][4][6]. Aberrant expression or mutation of CENP-H is implicated in cancer and may contribute to chromosomal instability syndromes[6][8]. No direct interacting drugs or mechanisms of action for small molecule targeting are currently described in the literature.
