Centromere protein O (CENPO) is a protein localized to the centromere throughout the cell cycle and is an essential component for proper chromosome segregation during mitosis[6]. It is a subunit of the CCAN (constitutive centromere-associated network), which interacts with centromeric chromatin to facilitate assembly and stabilization of the kinetochore complex[2]. CENPO is evolutionarily conserved and plays a critical role in ensuring genomic stability by participating in the assembly of the centromere-kinetochore structure, a prerequisite for accurate chromosome movement and segregation. Dysregulation or mutation of CENPO has been implicated in chromosomal instability found in cancer cells, hence its consideration as a potential biomarker and research target in oncology[6][2]. No approved drugs currently target CENPO directly.
