Centromere protein U (CENPU) is an essential, conserved centromere-associated factor required for the assembly and function of the kinetochore, a key structure for accurate chromosome segregation during mitosis[1][2][5]. CENPU functions within multiprotein centromere complexes, mediating kinetochore assembly, recruiting mitotic regulators (notably Polo-like kinase 1), directly interacting with core kinetochore components, and stabilizing kinetochore-microtubule attachment. It also plays a role in the spindle assembly checkpoint and may be involved in DNA double-strand break repair. Dysregulation or loss of CENPU leads to chromosome misalignment and segregation errors, contributing to conditions such as aneuploidy and has been implicated in cancer development[1][5].
