Centrosomal protein 128 kDa (CEP128) is a structural protein localized at the subdistal appendages of centrioles, where it is essential for centrosome integrity, microtubule organization, and ciliary vesicle trafficking[1][3]. It plays a key regulatory role in transforming growth factor-β/bone morphogenetic protein (TGF-β/BMP) signaling, especially at the primary cilium, affecting cellular signaling and homeostasis[3]. CEP128 is necessary for proper spermatogenesis; mutations or loss of CEP128 cause severe male infertility in mice and are associated with sperm defects in humans[2][5]. Moreover, circular RNA isoforms from CEP128 (circ‑CEP128) act as competing endogenous RNAs, modulating microRNA pathways that influence cancer cell proliferation, migration, apoptosis, and chemoresistance[1]. There is no evidence that CEP128 is a therapeutic drug target or that it is directly modulated by existing drugs.
