Centrosomal protein 131 kDa (CEP131) is a centriolar satellite-associated protein that localizes near the centrosome and plays crucial roles in centrosome homeostasis, centriole duplication, and primary cilia formation. CEP131 acts as an interactor and stabilizer of the Plk4/STIL complex, modulating the recruitment and stabilization of factors essential for the initiation of centrosome duplication. Loss or dysregulation of CEP131 affects the dynamics of microtubules, delays mitochondrial-dependent apoptosis, alters proteostasis, and is linked to genomic instability. Overexpression of CEP131 promotes centrosome amplification, genomic aberrations, and accelerates tumor formation, identifying it as a potential therapeutic target, particularly in oncology.
