Overview

Centrosomal protein 170 (CEP170) is a structural protein of the centrosome, specifically localizing to the subdistal appendages of the mature mother centriole during interphase, and to spindle microtubules near centrosomes during mitosis[1][2][3][4]. This protein binds microtubules, facilitates their anchoring at the centrosome, plays a vital role in microtubule organization, and regulates aspects of cell morphology and centrosome positioning[1][2][4][6]. CEP170 is phosphorylated by polo-like kinase 1 (PLK1) and Cdk1, and interacts with other proteins including IFT81 and PRAX-1[1][2][3][4][6]. It is essential for the assembly of subdistal appendages and is involved in cilium disassembly, centriole maturation, and possibly regulation of cell cycle progression[1][2][4]. Current research links CEP170 to rare genetic diseases (such as primary microcephaly), but it is not considered a direct therapeutic target or druggable molecule, nor is it established as a disease biomarker or safety liability at present[3]. Experimental overexpression or depletion alters cell shape and causes cytoskeletal defects without grossly disrupting centrosome structure, highlighting a specialized organizational role rather than an enzymatic or receptor-mediated function[1][4][6].
