Overview

Centrosomal protein 350 (CEP350) is a large, centrosome-localized structural protein essential for the proper organization and anchoring of microtubules at the centrosome, the principal microtubule-organizing center in animal cells[1][3]. The protein contains a CAP-Gly domain typical of cytoskeleton-associated proteins and forms a complex with FOP, another centrosomal component, to enable efficient microtubule anchoring, contributing to the integrity and organization of the interphase microtubule network[1][3]. CEP350 is also crucial for centriole growth by stabilizing procentriolar components, supports cilium formation (ciliogenesis), and directly regulates subcellular localization and activity of nuclear hormone receptors, such as PPARA, by recruiting them to specific compartments[1][2]. CEP350 interacts with other centrosomal proteins (e.g., CEP78, FOP/FGFR1 oncogene partner), playing a role in recruiting and stabilizing these proteins at the centrosome, and thus maintaining the functional microtubule network and centrosome structure[2][3]. Pathogenic mutations or knockdown of CEP350 can disrupt cilium assembly and cell division and may contribute to rare syndromes involving epidermal appendages[1]. However, no approved or experimental drugs currently target CEP350, nor is it considered a therapeutic drug target[1].
