Overview

Centrosomal protein 57 like 1 (CEP57L1) is a centrosome-associated protein involved in the maintenance of centriole engagement and proper centrosome duplication during the cell cycle[2][5][3]. It cooperates with its paralog CEP57 to ensure that centrioles remain properly engaged during interphase, preventing premature centriole disengagement and reduplication[2]. Loss or depletion of CEP57L1, especially in combination with loss of CEP57, results in abnormal centriole disengagement, centriole reduplication, and chromosome segregation errors that can contribute to cell division defects and potentially to genomic instability in cancer[2]. CEP57L1 is predicted to bind identical proteins and gamma-tubulin[3][6], is located at the centrosome, and is required for microtubule attachment to centrosomes, impacting proper organization and tethering of microtubule minus ends[7]. However, it is not regarded as a classic therapeutic target such as a receptor, enzyme, or transporter.
