Overview

Centrosomal protein 63 (CEP63) is a scaffold protein localized at the centrosome, the cell's main microtubule-organizing center[1][3]. It contains six coiled-coil domains and plays a central role in mother-centriole-dependent centriole duplication and spindle assembly, which are essential for proper cell division and genome stability[1][2][5]. CEP63 acts through direct interactions with other centrosomal proteins, notably CEP152, which together form a complex required for recruiting components necessary for procentriole formation and centriole-specific pathways[2][5]. Mutations in CEP63 are associated with human diseases including Seckel syndrome type 6 (a primary microcephaly disorder), myofibrillar myopathy 7, and have been implicated in certain cancers such as papillary thyroid cancer and colorectal cancer through promoting cell proliferation, migration, and survival[1][6][7]. CEP63 also participates in DNA damage response; its removal from centrosomes after DNA damage inactivates spindle assembly and leads to cell cycle delay[1]. There are currently no drugs reported to specifically target CEP63, nor established clinical biomarker or safety concerns linked to direct therapeutic targeting.
