Centrosomal protein of 295 kDa (CEP295) is a conserved scaffold protein that plays a pivotal role in the maturation and conversion of centrioles into fully competent centrosomes, which are essential for accurate cell division. CEP295 is recruited early during procentriole assembly to the proximal centriole wall and acts as a bridge, linking inner centriolar components with outer elements required for pericentriolar material (PCM) recruitment and establishing microtubule-organizing center activity. It is essential for centriole elongation, stability, and the recruitment of proteins such as POC5 and POC1B, as well as for post-translational modifications of centriolar microtubules. CEP295 also regulates cell proliferation and migration and has been implicated as a potential oncogenic factor in cancer, notably hepatocellular carcinoma. Its loss impairs centriole assembly, cell proliferation, and centrosome function, and it is associated with genetic disorders such as microcephaly and Seckel syndrome 11[1][2][3][5][6][7][9].
