Overview

Centrosomal protein of 44 kDa (CEP44) is a conserved, non-enzymatic structural protein localized to the proximal end and lumen of centrioles within the centrosome[1][2][3]. CEP44 is essential for centrosome cohesion (holding duplicated centrosomes together until their separation in mitosis), ensuring accurate centriole duplication, and enabling the centriole-to-centrosome conversion required for the formation of mature centrosomes capable of nucleating microtubules[1][2][3]. CEP44 closely interacts with structural proteins such as rootletin and POC1B, stabilizing these components for proper centrosome function[1][3]. Loss of CEP44 disrupts centrosome cohesion, centriole structure, and mitotic spindle pole formation, leading to defects in cell division[1][2][3]. Expression of CEP44 is downregulated in breast carcinoma, correlating with poorer clinical outcomes, which suggests a potential (though as yet non-therapeutic) role in tumor suppression mechanisms[2]. There are no known drugs that directly target CEP44, and it is not currently classified as a therapeutic target.
