Overview

Centrosomal protein of 72 kDa (CEP72) is a member of the leucine-rich-repeat (LRR) superfamily and localizes to the centrosome, a non-membrane-bound organelle that serves as the major microtubule-organizing center in animal cells[3]. CEP72 functions as a centriolar satellite/pericentriolar material protein that is essential for recruitment and organization of key components for centrosome assembly, including γ-tubulin ring complexes, AKAP9, KIZ, and proteins involved in centriole duplication[1][3]. Its depletion impairs microtubule nucleation, disrupts spindle integrity, and affects chromosome alignment during mitosis; in mice, loss of Cep72 can cause abnormal sperm morphology but does not impair fertility[1][4]. There are currently no known drugs that directly target CEP72 and it is not a validated therapeutic target, but it plays a crucial role in fundamental processes such as cell division and may be implicated in certain disease contexts[3][4].
