Centruroides scorpion venom neurotoxins are a group of potent, basic peptides found in the venom of scorpions from the Centruroides genus, such as the Arizona bark scorpion (Centruroides sculpturatus). These toxins primarily target voltage-gated sodium channels (Nav) in the nervous system, where they act as either alpha-toxins that slow channel inactivation or beta-toxins that shift the threshold of activation to more negative potentials (PubMed, PMID: 22107104). This physiological interference leads to persistent depolarization and the repetitive firing of action potentials, resulting in a massive release of neurotransmitters known as an autonomic storm. Clinically, envenomation manifests as severe neuromuscular hyperactivity, cranial nerve dysfunction, and potentially fatal respiratory distress (NIH, 2021). In a therapeutic context, these neurotoxins are the direct target of antivenom treatments like Anascorp, which utilize F(ab')2 antibody fragments to sequester the toxins in the vascular compartment and neutralize their activity. Beyond their role in pathology, these toxins are valuable tools in neurobiology for studying the structure and gating mechanisms of ion channels (UniProt, 2023).
