Ceramide 1, also known as Ceramide EOS, is a critical sphingolipid component of the mammalian skin barrier, specifically the stratum corneum [31]. It is characterized by an omega-hydroxy fatty acid that is ester-linked to another fatty acid (typically linoleic acid) and amide-linked to a sphingosine base [31]. This unique structure allows Ceramide 1 to act as a molecular rivet, bridging lipid lamellae and maintaining the structural integrity of the skin's permeability barrier [31]. Deficiencies in Ceramide 1 are strongly associated with skin disorders such as atopic dermatitis, psoriasis, and ichthyosis [24]. While not a traditional therapeutic target like a receptor or enzyme, it is a key active ingredient in dermatological treatments aimed at restoring skin barrier function and preventing transepidermal water loss [31]. In signaling contexts, the term is sometimes confused with Ceramide-1-phosphate (C1P), a bioactive lipid that regulates cell survival and inflammation through a putative G protein-coupled receptor [1, 2].
