Ceramide-1-phosphate transfer protein (CPTP) is a ubiquitously expressed human lipid transfer protein encoded by GLTPD1, dedicated to shuttling the bioactive sphingolipid ceramide-1-phosphate (C1P) between organelle and plasma membranes in the cytosol. Structurally, CPTP uses a two-layer, all-alpha-helical GLTP fold and features a unique surface-phosphate recognition site that confers specificity for C1P, facilitating a “hydrocarbon chain-first” mechanism for lipid uptake and release. Functionally, CPTP maintains proper sphingolipid homeostasis by regulating C1P distribution, which is essential for inflammatory signaling, programmed cell death, and eicosanoid production through activation of cytosolic phospholipase A2. Deficiency or overactivity of CPTP alters membrane architecture, triggers excessive autophagy and inflammasome activity, and increases the release of pro-inflammatory cytokines such as interleukin-1β and interleukin-18. CPTP’s modulation of these key processes links it to pathological inflammation and suggests potential disease relevance in conditions requiring tight regulation of cell death and immune responses. No drugs directly targeting CPTP are currently established. If further structured or drug-related data are needed, additional research and database integration will be required.
