Overview

Cerebellar Purkinje cells are a class of large GABAergic neurons located in the cerebellum and are characterized by their elaborate dendritic arborization. They serve as the primary integration center and the sole inhibitory output of the cerebellar cortex, playing a fundamental role in motor control, coordination, and precision (StatPearls, 2023). These cells receive excitatory inputs from parallel fibers and climbing fibers, which they process to regulate motor learning and timing (NIH, 2022). Degeneration or loss of Purkinje cells is a central feature in many neurodegenerative diseases, including spinocerebellar ataxias (SCAs) and gluten ataxia (PubMed, 2021). Although the cell itself is not a single molecular target, its specific ion channels (such as P/Q-type calcium channels) and receptors (such as mGluR1) are major sites for pharmacological intervention in movement disorders (UniProt, 2023). Drugs like riluzole are often investigated for their ability to modulate the excitability of these neurons to alleviate symptoms of cerebellar dysfunction (PubMed, 2020).
