Overview

Cerebellin-1 precursor protein (CBLN1) is a secreted glycoprotein specifically enriched in the cerebellum but also found in various other brain regions and neuroendocrine tissues. It is processed proteolytically to give rise to smaller peptides, including cerebellin, which is highly concentrated in postsynaptic structures of Purkinje cells. Functionally, CBLN1 is essential for the integrity, differentiation, and maintenance of both excitatory and inhibitory synapses, primarily by forming trans-synaptic complexes that bridge presynaptic neurexins and postsynaptic GluD2 glutamate receptor subunits. CBLN1 also plays roles in axon guidance during neuronal development and influences certain endocrine processes, such as modulating corticosteroid secretion and insulin production. Mutations or dysregulation can result in severe motor coordination and cognitive impairments, as observed in certain neurodevelopmental syndromes. Despite its pivotal roles in neuronal physiology and development, it is not currently a therapeutic drug target.
