Overview

CGG triplet repeat-binding protein 1 (CGGBP1) is a nuclear protein encoded by the CGGBP1 gene on chromosome 3p11.1 in humans. It specifically binds to unmethylated CGG trinucleotide repeats, such as those in the promoter of the FMR1 gene, and plays a role in regulating transcription, repressing elements such as Alu repeats, and modulating gene expression by binding to promoter regions containing CGG repeats[1][3][4][7]. It is essential for cell survival due to its roles in DNA repair, telomere maintenance, and general cytoprotection[1][4]. CGGBP1 may affect chromatin barrier functions and the pattern of CTCF (CCCTC-binding factor) binding, influencing higher-order genome organization[6]. Although CGGBP1 binds CGG repeats in the FMR1 gene and its binding is methylation-sensitive, functional studies indicate it does not directly regulate FMR1 transcription or CGG expansion stability but may act as a modulatory or structural accessory factor in transcriptional regulation[3][4][7]. There is no current evidence that CGGBP1 is a direct therapeutic target, nor are drugs, mechanisms of action, biomarkers, or safety concerns associated with targeting this protein reported in available literature.
