cGMP-dependent protein kinase I alpha (PKG Iα) is a serine/threonine protein kinase that serves as a key mediator of the nitric oxide (NO)/cyclic guanosine monophosphate (cGMP) signaling pathway. It plays central roles in multiple physiological processes including relaxation of vascular and gastrointestinal smooth muscle, inhibition of platelet aggregation, modulation of cardiac hypertrophy, and neuroprotection. Recent research also implicates nociceptor-localized PKGI as critical for central sensitization underlying neuropathic pain states.
