The cGMP-enzyme regulatory system is a network of enzymes and signaling molecules that generate, degrade, and respond to cyclic guanosine monophosphate (cGMP), a key second messenger involved in numerous physiological processes. It includes guanylate cyclases (sGC and pGC), phosphodiesterases (PDEs), and cGMP-regulated effectors like Protein Kinase G (PKG) and cyclic nucleotide-gated ion channels. This system regulates diverse functions such as vascular tone, cardiac function, neurotransmission, phototransduction, and more. Dysregulation can contribute to various diseases, making it a target for therapeutic interventions.
Nitrates increase NO production, leading to sGC activation and increased intracellular cGMP. Phosphodiesterase inhibitors prevent the breakdown of intracellular cGMP.
