Overview

Charged multivesicular body protein 1A (CHMP1A) is a multifunctional protein encoded by the CHMP1A gene. It belongs to the ESCRT-III complex, essential for sorting proteins into multivesicular bodies (MVBs) for lysosomal degradation, which is a crucial step in membrane remodeling during cell division (cytokinesis) and viral budding. Besides its cytoplasmic functions, CHMP1A acts within the nucleus to regulate chromatin organization, gene expression, and cell cycle progression by interacting with Polycomb-group proteins (such as BMI1) and regulating ATM and p53 signaling pathways. CHMP1A has a documented role as a tumor suppressor in pancreatic and renal cancers, and its disruption can lead to neurodevelopmental diseases like pontocerebellar hypoplasia. Mechanism of action

Drugs or agents that modulate CHMP1A would affect ESCRT-III complex function, membrane scission, lysosomal protein degradation, cell cycle arrest, and tumor suppression signaling (via ATM and p53).
