Charged multivesicular body protein 3 (CHMP3) is a core component of the endosomal sorting complex required for transport-III (ESCRT-III), which is involved in the formation of multivesicular bodies (MVBs) and in sorting endosomal membrane proteins into lysosomes for degradation[1][5][6]. CHMP3 participates in membrane budding and scission events, recruits cofactors for ESCRT-III assembly (such as VPS4), and plays roles in processes including cytokinesis, the terminal stage of cell division, and the budding of some enveloped viruses such as HIV-1[1][2][5]. CHMP3 helps regulate protein trafficking, degradation of growth factor receptors (like EGFR), and cellular stress responses. It is implicated in key mechanisms of cancer cell proliferation, migration, invasion, and in maintaining normal neuroendocrine cell differentiation[5][8]. It has been studied in diseases such as hepatocellular carcinoma, frontotemporal dementia/ALS, and triple-negative breast cancer, where its expression influences tumor cell behavior and patient outcomes[5][8]. There are currently no known drugs that directly target CHMP3.
