Charged multivesicular body protein 6 (CHMP6) is a core component of the ESCRT-III complex, encoded by the human CHMP6 gene located on chromosome 17. CHMP6 mediates the formation of multivesicular bodies (MVBs) by facilitating the sorting of endosomal cargo into intraluminal vesicles, which are subsequently delivered to lysosomes for degradation of membrane proteins, lipids, and lysosomal enzymes. It acts as an acceptor for the ESCRT-II complex at the endosomal membrane, and forms part of the machinery for membrane fission events, including cytokinesis and viral budding (HIV-1 and other lentiviruses release). CHMP6 is myristoylated at the N-terminus and interacts with other ESCRT-III subunits (such as CHMP4b) and auxiliary components (like VPS4 ATPase). Mutations or dysregulation of CHMP6 have been linked to neurodegenerative disorders (frontotemporal dementia and ALS) and viral infectious disease pathways. It is not considered a classical drug target, biomarker, or safety concern at this time.
