Chemerin chemokine-like receptor 2 (CMKLR2, also known as CCRL2) is an atypical seven-transmembrane G protein-coupled receptor that binds the chemoattractant protein chemerin at its N-terminus but does not trigger canonical intracellular signaling or cell migration typically associated with GPCRs[1][3][5]. Unlike the functional signaling chemerin receptor CMKLR1, CCRL2 primarily serves as a non-signaling cell-surface anchoring protein that presents chemerin to other chemerin-responsive cells, especially at barrier tissues such as endothelial cells[1]. This receptor does not appreciably internalize or scavenge its ligand under physiological conditions and is believed to fine-tune immune responses by regulating chemerin’s local availability and presentation. Although implicated in the modulation of inflammation and immune cell trafficking, CCRL2 is currently viewed as a non-classical, non-signaling receptor, with disease roles and therapeutic targeting still under investigation[1][5].
