Overview

Chemokine-like receptor 1 (CMKLR1), also known as ChemR23 or chemerin receptor 23, is a G protein-coupled receptor (GPCR). It primarily binds the adipokine chemerin and the specialized pro-resolving lipid mediator resolvin E1. CMKLR1 is expressed by immune cells (macrophages, dendritic cells, NK cells, microglia), cardiomyocytes, adipocytes, and endothelial cells. Activation of CMKLR1 mobilizes intracellular calcium and triggers signaling pathways. It is involved in chemoattraction of immune cells, inflammation modulation, metabolic regulation, and angiogenesis. Targeting CMKLR1 has therapeutic potential in neuroinflammatory diseases and metabolic disorders.

Mechanism of action

Activation of CMKLR1 mobilizes intracellular calcium and triggers signaling pathways including ERK1/2 and NF-kB. Chemerin/CMKLR1 interaction promotes migration of immune cells and may also help resolve inflammation through enhanced efferocytosis. Resolvin E1 binding to CMKLR1 suppresses leukocyte infiltration during inflammatory responses.
