Chemical modifier is a broad, non-specific term in pharmacology and toxicology used to describe substances that alter the biological response to a therapeutic agent, environmental stressor, or physiological process. In clinical oncology, the term most frequently refers to 'chemical modifiers of radiation response,' which include radiosensitizers like misonidazole that enhance the lethal effects of radiation on tumor cells and radioprotectors like amifostine that safeguard normal tissues (Oncohema Key, 2016). Additionally, the term encompasses 'chemical genetic modifiers' used in phenotype-based screenings to identify or perturb signaling pathways in model organisms like zebrafish (Longdom, 2013; PMC, 2009). Other applications include modifiers of drug delivery, such as polylysine, which enhance the transport of pharmaceutical agents (Google Patents, 1993). Because it denotes a functional category or a methodological approach rather than a specific biological target such as a distinct protein, enzyme, or receptor, 'Chemical modifier' is considered an incorrect or overly generic entry for a therapeutic target (NCI Thesaurus C1908).
Mechanisms vary by agent; radiosensitizers often act as oxygen mimetics to stabilize radiation-induced DNA damage in hypoxic cells, whereas radioprotectors typically act as free radical scavengers to shield healthy tissue. Other chemical modifiers may allosterically regulate proteins, such as hemoglobin, to increase oxygen delivery to tissues.
