Overview

Chemoattractant Receptor Homologous Molecule Expressed on Th2 Cells (CRTH2), also known as Prostaglandin D2 receptor 2 (DP2), is a G protein-coupled receptor that mediates the chemotaxis of Th2 lymphocytes, eosinophils, and basophils toward inflammatory sites upon binding its primary ligand Prostaglandin D2 (PGD2). Activation leads to increased intracellular calcium levels, reduced cyclic AMP levels, and activation of signaling pathways, resulting in recruitment and activation of Th2-type immune cells during allergic inflammation. CRTH2 plays a central role in mediating allergic inflammation and is a therapeutic target for diseases like severe eosinophilic asthma. Antagonists targeting CRTH2 are under clinical development.
