Overview

Chemokine-like factor 1 (CKLF1) is a secreted cytokine belonging to the chemokine family, encoded by the CKLF gene on chromosome 16q22.1. Four isoforms exist (CKLF1-4) due to alternative splicing, with CKLF1 being the most studied. CKLF1 is a potent chemoattractant for neutrophils, lymphocytes, and monocytes, promoting cell migration, proliferation, and differentiation. It acts mainly through the CCR4 receptor, a G protein-coupled receptor on immune cells. CKLF1 is implicated in diverse physiological and pathological processes including immune regulation, inflammation, tissue regeneration, cardiovascular disease, cancer, and reproductive function. Though no marketed drugs directly target CKLF1, experimental antibodies and antagonists are being explored, with CKLF1 considered a promising therapeutic target especially for inflammatory diseases and cancer.
