The CMTM6–PD-L1 complex is a membrane-associated protein complex formed by the interaction of CMTM6 (chemokine-like factor MARVEL transmembrane domain–containing protein 6) with programmed cell death ligand 1 (PD-L1). CMTM6 physically associates with PD-L1 at the plasma membrane and recycling endosomes, protecting PD-L1 from lysosomal and proteasomal degradation and enhancing its half-life at the cell surface[1][2][3][4][5]. This stabilization enables PD-L1 to effectively suppress T-cell–mediated immune responses by binding to PD-1, allowing tumor cells to evade immune attack. The CMTM6–PD-L1 complex is considered a therapeutic target in cancer immunotherapy, as disrupting their interaction can decrease immune evasion and promote anti-tumor immunity. Expression levels of both CMTM6 and PD-L1 are positively correlated with poor prognosis in multiple tumors, and they serve as potential biomarkers for patient selection and monitoring in immune checkpoint blockade therapies[1][2][5].
