Overview

Chemokine production is the biological process by which various cell types, including macrophages, fibroblasts, and endothelial cells, synthesize and release chemotactic cytokines (chemokines) in response to physiological or pathological stimuli [1, 6]. These small proteins establish chemical gradients that guide the migration and activation of leukocytes to sites of infection, injury, or inflammation [2, 9]. While essential for normal immune surveillance and homeostatic tissue maintenance, the dysregulation of this process is a key driver of chronic inflammatory conditions, such as rheumatoid arthritis and multiple sclerosis, as well as the progression and metastasis of various cancers [2, 7, 12]. Therapeutic strategies often aim to curb excessive chemokine production by targeting upstream signaling pathways, such as NF-κB, MAP kinase, or JAK/STAT, or by blocking the primary cytokines like TNF-α and IL-1 that induce chemokine expression [5, 11, 19]. As such, chemokine production represents a critical pathological endpoint and a readout for drug efficacy rather than a single discrete molecular target [14, 18].
