Overview

Chicken ovalbumin upstream promoter-transcription factor II (COUP-TFII), also known as NR2F2, is a member of the orphan nuclear receptor superfamily that functions as a key transcriptional regulator [UniProt P24466, NCBI Gene 7026]. It plays a critical role in embryonic development, particularly in the specification of venous identity by repressing arterial genes, and is essential for heart morphogenesis and the development of the lymphatic system [Nature 2005, Nature 2010]. In adults, COUP-TFII is involved in maintaining metabolic homeostasis, regulating glucose levels, and modulating circadian rhythms [JBC 2007]. Pathologically, it is frequently overexpressed in various malignancies, including prostate and colorectal cancers, where it promotes tumor progression, angiogenesis, and metastasis by modulating the tumor microenvironment and epithelial-mesenchymal transition [Nature Comm 2013, Cancer Res 2015]. While it is considered a high-priority therapeutic target due to its role in cancer and metabolic diseases, the development of specific small-molecule modulators remains a challenge because its natural ligand is not well-defined, though it has shown low-affinity binding to certain retinoids [PLOS Bio 2008].
