Overview

The Chikungunya virus E1 envelope protein is a critical structural component of the Chikungunya virus (CHIKV), an alphavirus transmitted by Aedes mosquitoes. It functions as a class II viral fusion protein, playing a primary role in the fusion of the viral envelope with the host endosomal membrane following receptor-mediated endocytosis [1][2]. E1 exists as a heterodimer with the E2 glycoprotein on the viral surface; while E2 facilitates initial attachment to host receptors, E1 mediates the low-pH-triggered conformational change necessary for membrane penetration [3]. This protein is a major target for the host immune response and a focal point for the development of neutralizing monoclonal antibodies and fusion inhibitors [4]. Mutations in the E1 protein, such as the A226V substitution, have historically influenced the virus's vector specificity and epidemic potential [5]. Therapeutic strategies targeting E1 aim to block the fusion process or prevent the structural transitions required for viral entry into target cells [6].
