Chimeric antigen receptor targeting CD70 refers to engineered T-cell therapies, typically allogeneic CAR T cells, designed to recognize and bind the CD70 antigen on cancer cells, triggering immune-mediated destruction. CD70 is a surface protein (CD27 ligand) overexpressed in various malignancies including T-cell lymphoma, acute myeloid leukemia (AML), clear cell renal cell carcinoma (ccRCC), and relapsed/refractory hematological cancers. These CARs are produced by modifying donor or patient T cells (e.g., via CRISPR-Cas9 for CTX130) to express an anti-CD70 receptor, often with edits to reduce rejection and fratricide. Clinical trials show promising objective response rates (e.g., 46-52% ORR in lymphoma, disease control in 81% of ccRCC), manageable safety profiles, and activity in CD70-positive tumors refractory to standard therapies
