Overview

Chimerin 2 (CHN2) is a Rho GTPase-activating protein (GAP) encoded by the CHN2 gene in humans[1][2][3]. It selectively acts on Rac1, a small GTPase involved in cytoskeletal regulation, and plays crucial roles in neural circuit development (such as axon pruning), as well as in cell proliferation and migration[1][2][3][5]. The protein contains several functional domains, including a phorbol-ester/diacylglycerol-type zinc finger, a Rho-GAP domain, and an SH2 domain, which allow it to be regulated by second messengers like diacylglycerol (DAG). CHN2 expression is linked to various pathologies, including certain cancers, neurodegenerative and psychiatric diseases, and metabolic disorders. Its ability to modulate Rac1-dependent signaling is central to its biological and disease-related functions[1][2][3][5].

Mechanism of action

There are currently no approved drugs known to target Chimerin 2 directly. The key mechanism is GTPase-activating protein (GAP) activity acting on Rac1, inactivating it by stimulating the hydrolysis of GTP to GDP.
