Chitin synthase 1 is a membrane-integrated glycosyltransferase enzyme responsible for the biosynthesis of chitin, a β-1,4-linked N-acetylglucosamine polymer crucial for cell wall structure in fungi and exoskeleton formation in arthropods[1][2][3][6][8]. In fungi such as Saccharomyces cerevisiae, Chitin synthase 1 (Chs1) acts as an auxiliary enzyme, particularly involved in repairing cell wall damage rather than primary septum formation, collaborating with other chitin synthases for overall cell wall integrity[5][6]. The enzyme operates by transferring GlcNAc residues from UDP-GlcNAc to the non-reducing end of a growing chitin chain, with the active site and polysaccharide transport channel located within its multi-transmembrane structure[1][2][3][6]. Inhibition of Chs1 by agents such as nikkomycin Z blocks chitin synthesis, causing cell wall defects and fungal cell death, making it a validated therapeutic target for antifungal drug development[1][2][3]. Since chitin is absent in mammals, selective inhibition of Chs1 is being explored for antifungal therapy with reduced host toxicity[2][3][8]. Chs1 shares homology and mechanistic features with cellulose and hyaluronan synthases but is functionally specialized for fungal and invertebrate chitin production[2][3].
