Overview

Chloride channel protein type 2 subunit alpha (ClC-2), encoded by the CLCN2 gene, is a voltage-gated and osmosensitive chloride channel expressed in various tissues, including enterocytes. It plays a critical role in maintaining chloride homeostasis, regulating cell volume, and influencing membrane potential. In intestinal epithelial cells, ClC-2 contributes to transepithelial ion transport and fluid secretion/absorption. Dysfunctions in CLCN2 have been linked to potential associations with neurological conditions such as epilepsy and leukoencephalopathy, although the clinical significance remains under investigation. Classical chloride channel blockers show weak inhibition; Zn²⁺ can inhibit activity experimentally.
