Chloride intracellular channel protein 3 (CLIC3) is an atypical chloride ion channel that can exist both as a soluble protein with glutaredoxin-like enzyme activity and, upon oxidative or pH triggers, insert into intracellular membranes to form ion channels[1][2][5]. It is localized mainly in the nucleus and other organelles but can also appear at the plasma membrane. CLIC3 is involved in regulating chloride ion fluxes, redox reactions, and extracellular matrix remodeling. Research implicates it in promoting angiogenesis in tumors, regulating immune responses, and playing a role in cellular growth, as well as pathological conditions such as cancer, cardiovascular, and pregnancy-associated diseases[1][2]. While not a major drug target as of 2024, its multifaceted cellular roles and disease associations are under active investigation.
