Choline kinase (CK, also known as ChoK) is a cytosolic enzyme that catalyzes the phosphorylation of choline to form phosphocholine in the presence of ATP and magnesium. This reaction is the first and committed step in the CDP-choline pathway for phosphatidylcholine (PC) biosynthesis, which is essential for membrane formation and cellular signaling. Overexpression or dysregulation of CK—especially ChoKα—is linked to malignant transformation across various cancers, making it a promising target for anticancer therapy. ChoKα has also been implicated as a potential therapeutic target for obesity.
Small molecule inhibitors block catalytic site or disrupt dimerization, decreasing phosphocholine levels and impairing tumor cell proliferation.
