Overview

Choline transporter-like protein 2 (SLC44A2) is a multi-pass transmembrane transporter and a member of the solute carrier 44 family, with high expression in platelets, red blood cells, brain endothelium, and specific cancer types[1][2][5]. Functionally, it is involved in the mitochondrial transport of choline, supporting mitochondrial ATP and ROS (reactive oxygen species) production, which is essential for platelet activation and aggregation, and thus plays a key role in hemostasis and thrombosis[2][3][4]. SLC44A2 is also critical for hearing preservation and the maintenance of cerebral vascular homeostasis; biallelic deficiency leads to progressive hearing loss and vascular abnormalities in humans[5]. Genetic studies have linked SLC44A2 variants to venous thromboembolism and the rare blood group antigens RIF and VER[5]. While not currently a direct therapeutic drug target, its role in platelet metabolism and thrombosis makes it a potential future target for antithrombotic therapy or for diseases related to mitochondrial dysfunction in hematopoietic and neural tissues[2][5].

Mechanism of action

In animal studies, choline transport via SLC44A2 supports mitochondrial metabolism, ATP and ROS production in platelets, thus facilitating platelet activation and thrombosis; loss of SLC44A2 impairs these processes, leading to reduced thrombosis[2][3][5]
