Choline trimethylamine-lyase (CutC)

Target
CutC
Molecular classification
Enzyme, Glycyl radical enzyme
01

Overview

Choline trimethylamine-lyase (CutC) is an anaerobic bacterial enzyme that catalyzes the cleavage of choline into trimethylamine (TMA) and acetaldehyde. This reaction is a key step in the microbial metabolism of choline within the human gut microbiota. The enzyme belongs to the glycyl radical enzyme (GRE) family and requires activation by a partner protein, CutD. TMA produced by this pathway is absorbed and converted in the liver to trimethylamine N-oxide (TMAO), which has been linked to increased risk of cardiovascular diseases and other metabolic disorders in humans. Inhibition of CutC is being explored as a therapeutic strategy.
