Overview

Chondroadherin is a small leucine-rich repeat proteoglycan (family: SLRP class IV) that is highly expressed in cartilaginous tissues such as articular cartilage and intervertebral discs[1][3]. It is characterized by 11 leucine-rich repeats flanked by cysteine-rich domains. Chondroadherin mediates specific adhesion of chondrocytes, fibroblasts, and osteoblasts primarily via binding to integrin α2β1 and syndecan cell-surface receptors, triggering activation of MAPK/ERK signaling pathways that help maintain the differentiated chondrocyte phenotype and cartilage homeostasis[1][2][3][5]. Chondroadherin plays a critical role in the organization of the extracellular matrix, particularly in the formation of the collagen fibrillar network required for the biomechanical strength of cartilage[1][3]. Genetic and proteomic studies suggest that alterations or fragmentation of chondroadherin are linked to degenerative changes in musculoskeletal tissues, serving as a sensitive biomarker for early degeneration and abnormal mechanical stress responses[1][4]. Although chondroadherin itself is not a direct therapeutic target, its modulation may have indirect therapeutic implications in cartilage repair, tissue engineering, and musculoskeletal disease detection[1][3][5].
