Overview

Chondroitin sulfate is a highly variable sulfated glycosaminoglycan composed of repeating disaccharide units—D-glucuronic acid and N-acetyl-D-galactosamine—often attached to core proteins forming proteoglycans within the extracellular matrix. It plays critical roles in providing structural integrity to cartilage by resisting compressive forces through osmotic swelling pressure. Beyond its mechanical functions, chondroitin sulfate interacts with numerous signaling molecules—including growth factors and cytokines—to regulate cellular behaviors such as proliferation and differentiation. Its diverse biological activities depend heavily on chain length and specific patterns of sulfation modifications. Abnormalities in its synthesis or modification are implicated in skeletal disorders like chondrodysplasia; altered interactions also contribute to diseases such as osteoarthritis and cardiovascular disease through effects on lipid retention within vessel walls.
