Overview

Chondroitin sulfate proteoglycan 4 (CSPG4) is a large, single-pass transmembrane proteoglycan characterized by an extensive extracellular domain decorated with chondroitin sulfate side chains, a transmembrane region, and a short intracellular tail. Originally described as a melanoma-associated antigen (MCSP or HMW-MAA), CSPG4 is highly expressed on various tumor cells—including melanoma, glioblastoma, sarcoma, and triple-negative breast cancer—while showing limited expression in normal adult tissues. CSPG4 lacks intrinsic kinase or catalytic activity, but acts as a scaffold, interacting with growth factors, integrins, matrix proteins, and receptor tyrosine kinases to promote activation of downstream oncogenic signaling cascades (notably MAPK and PI3K/AKT), thereby supporting tumor proliferation, migration, adhesion, ECM remodeling, and metastasis. Its restricted normal tissue distribution and key role in tumor biology make CSPG4 a prioritized target for therapeutic strategies such as monoclonal antibodies and CAR T-cell therapies in oncology.
