Overview

Chondroitin sulfate synthase 3 (CHSY3) is a glycosyltransferase enzyme involved in the biosynthesis of chondroitin sulfate, a major extracellular matrix glycosaminoglycan found in connective tissues. CHSY3 catalyzes the alternating transfer of glucuronic acid and N-acetylgalactosamine residues to the growing chondroitin polymer. It exhibits both beta-1,3-glucuronic acid and beta-1,4-N-acetylgalactosamine transferase activities, though with reduced activity compared to its paralog CHSY1. CHSY3 is implicated in tumorigenesis, as its overexpression promotes proliferation, migration, and invasion of gastric cancer cells and correlates with poorer patient prognosis across several tumor types. Additionally, CHSY3 expression is associated with extracellular matrix remodeling and greater immune infiltration in the tumor microenvironment, and high levels may predict resistance to immunotherapy in cancer settings. No approved drugs are known to specifically target CHSY3, but it is being explored as a prognostic biomarker and potential pharmacological target in oncology.
