Chordin-like protein 2 (CHRDL2)

Target
CHRDL2
Molecular classification
Secreted protein, Extracellular matrix-associated protein, Cysteine-rich domain-containing protein, BMP antagonist family
01

Overview

Chordin-like protein 2 (CHRDL2) is a secreted, cysteine-rich protein sharing structural similarity with chordin family members, including a pro-collagen repeat domain. CHRDL2 directly interacts with activin A and is expressed in various tissues. Notably, its differential expression in osteoblasts and increased expression in osteoarthritic cartilage point to roles in skeletogenesis and cartilage regeneration. Mutations or dysregulation have been linked to diseases such as inflammatory bowel disease and agammaglobulinemia[3].

Chordin-like 2CHRDL2
02

Mechanism of action

Binds and modulates activity of human activin A, potentially influencing TGF-β pathway signaling[3].

03

Biological functions

Directly binds human activin A (a member of the TGF-β superfamily)Modulates growth factor signalingDifferential expression during osteoblast differentiationRole in adult cartilage regeneration (upregulated in osteoarthritic joint cartilage)
04

Disease associations

Inflammatory bowel diseaseAgammaglobulinemiaOsteoarthritis (expression upregulated in osteoarthritic cartilage)
05

Safety considerations

None specifically described in current literature. General safety concerns may arise from modulating growth factor pathways.
06

Biomarkers

Upregulation in osteoarthritic cartilage suggests CHRDL2 expression as a potential biomarker for cartilage regeneration and osteoarthritis[3].
